Minisatellites, microsatellites, and short random oligonucleotides all uncover highly polymorphic DNA fingerprint patterns in Southern analysis of genomic DNA that has been digested with a restriction enzyme having a 4-bp specificity. The polymorphic nature of the fragments is attributed to tandem repeat number variation of embedded minisatellite sequences. This explains why DNA fingerprint fragments are uncovered by minisatellite probes, but does not explain how it is that they are also uncovered by microsatellite and random oligonucleotide probes. To clarify this phenomenon, we sequenced a large bovine genomic BamHI restriction fragment hybridizing to the Jeffreys 33.6 minisatellite probe and consisting of small and large Sau3A-resistant subfragments. The large Sau3A subfragment was found to have a complex architecture, consisting of two different minisatellites, flanked and separated by stretches of unique DNA. The three unique sequences were characterized by sequence simplicity, that is, a higher than chance occurrence of tandem or dispersed repetition of simple sequence motifs. This complex repetitive structure explains the absence of Sau3A restriction sites in the large Sau3A subfragment, yet provides this subfragment with the ability to hybridize to a variety of probe sequences. It is proposed that a large class of interspersed tracts sharing this complex yet simplified sequence structure is found in the genome. Each such tract would have a broad ability to hybridize to a variety of probes, yet would exhibit a dearth of restriction sites. For each restriction enzyme having 4-bp specificity, a subclass of such tracts, completely lacking the corresponding restriction sites, will be present. On digestion with the given restriction enzyme, each such tract would form a large fragment.(ABSTRACT TRUNCATED AT 250 WORDS)
We investigated the effects of feedback (grades) on two primary mood dimensions-Pleasantness and Arousal. We hypothesized that (a) cognitive comparisons of feedback to multiple standards yield cognitive appraisals of feedback sign, and that (b) feedback sign has a linear effect on harm/benefit appraisal, which influences mood Pleasantness, and a curvilinear effect on need for action appraisal, which influences Arousal. In Study 1 (N = 281), grades (a proxy for the sign of feedback-norm discrepancy) had a linear relationship with Pleasantness and a U-shaped relationship with Arousal. In addition, subjects who received grades had higher Arousal than control subjects. Study 2 (N = 226) replicated the results of Study 1 and showed that after controlling for the relationship of grades with Pleasantness and Arousal, the sign of feedback-expectation discrepancy (a second standard) also had a linear relationship with Pleasantness and a U-shaped relationship with Arousal. The role of large high-technology firms in fashioning the spatial extent of the labour markets that serve them, is examined in this paper. It is argued that the demand for highly skilled labour in these firms results in their active role in labour market extension through a strategy of employee transport. This makes for employment mobility without a commensurate impact on residential mobility. It can also result in the 'inclusion' or 'exclusion' of certain types of labour. In this context, the 'free-rider' phenomenon associated with worker transport is identified and described. On the basis of an empirical study of some of the largest high-technology firms in metropolitan areas in Israel, these processes are illustrated. The labour markets serving these firms are delimited and characterised for employees of different skill levels. In addition, the determinants of the probability of the firm utilising spatially extensive labour markets is examined. The role of firm size in explaining this behaviour is stressed.

A molecular, mean-field theory of chain packing statistics in aggregates of amphiphilic molecules is applied to calculate the conformational properties of the lipid chains comprising the hydrophobic cores of dipalmitoyl-phosphatidylcholine (DPPC), dioleoyl-phosphatidylcholine (DOPC), and palmitoyl-oleoyl-phosphatidylcholine (POPC) bilayers in their fluid state. The central quantity in this theory, the probability distribution of chain conformations, is evaluated by minimizing the free energy of the bilayer assuming only that the segment density within the hydrophobic region is uniform (liquidlike). Using this distribution we calculate chain conformational properties such as bond orientational order parameters and spatial distributions of the various chain segments. The lipid chains, both the saturated palmitoyl (-(CH2)(1)4-CH3) and the unsaturated oleoyl (-(CH2)(7)-CH=CH-(CH2)(7)-CH3) chains are modeled using rotational isomeric state schemes. All possible chain conformations are enumerated and their statistical weights are determined by the self-consistency equations expressing the condition of uniform density. The hydrophobic core of the DPPC bilayer is treated as composed of single (palmitoyl) chain amphiphiles, i.e., the interactions between chains originating from the same lipid headgroup are assumed to be the same as those between chains belonging to different molecules. Similarly, the DOPC system is treated as a bilayer of oleoyl chains. The POPC bilayer is modeled as an equimolar mixture of palmitoyl and oleoyl chains. Bond orientational order parameter profiles, and segment spatial distributions are calculated for the three systems above, for several values of the bilayer thickness (or, equivalently, average area/headgroup) chosen, where possible, so as to allow for comparisons with available experimental data and/or molecular dynamics simulations. In most cases the agreement between the mean-field calculations, which are relatively easy to perform, and the experimental and simulation data is very good, supporting their use as an efficient tool for analyzing a variety of systems subject to varying conditions (e.g., bilayers of different compositions or thicknesses at different temperatures).
Artificial selection for increased resistance to methidathion in two replicate lines of the whitefly, Bemisia tabaci (Gennadius) (originating from a cotton-field-collected greenhouse population) was successful; LC(50)s increased 7-8.6-fold in eight generations in the selected lines. This indicates the existence of additive genetic variance for resistance in the source population. Minimal realized heritability estimates, calculated from the response to selection, were h(2) = 0.49 after one generation (with minimal effects of common laboratory environment and inbreeding) and a mean value of h(2) = 0.344 after eight generations. Esterase activity (measured from the hydrolysis of beta-naphthyl butyrate) increased in the selected lines in correlation with resistance. We observed no change in mean esterase activity in the unselected (control) population. No consistent differences in fitness components between selected and control lines were detected during selection, but females exposed to sublethal doses of methidathion tended to have increased fecundity.
